The positive Phase 2 clinical trial results for trastuzumab pamirtecan are expected to positively impact BioNTech's stock price and market valuation, as it signifies a significant advancement in its oncology pipeline. This development also enhances the reputation of DualityBio, its co-developer, and could attract further investment in the biotechnology sector, particularly for companies focused on antibody-drug conjugates.
BioNTech announced positive results from a Phase 2 clinical trial for trastuzumab pamirtecan (BNT323/DB-1303), an investigational HER2-targeted antibody-drug conjugate, in patients with HER2-expressing, recurrent endometrial cancer. The trial demonstrated clinically meaningful antitumor activity across all HER2 expression levels and a manageable safety profile, with a confirmed objective response rate of 47.9% in centrally HER2-tested patients and a median progression-free survival of 8.1 months. This data supports the potential of trastuzumab pamirtecan in real-world patient populations, including those with lower HER2 expression levels and prior checkpoint inhibitor treatment. BioNTech and DualityBio plan to file a biologics license application in 2026, following previous Fast Track and Breakthrough Therapy designations from the United States — Food and Drug Administration. The drug is also being evaluated in two global Phase 3 clinical trials for endometrial cancer and metastatic breast cancer.
