Led by Seema Khurana, the team proposes a paradigm shift from symptom management to addressing inherent defects in the epithelial lining, which drives the disease. They found that in Crohn s disease patients, a chronic stress signal in epithelial cells leads to programmed cell death (necroptosis) and inhibits regeneration of the gut barrier. By repurposing two existing cancer medications, Pazopanib and Ponatinib, at low concentrations, the researchers were able to inhibit this stress response and promote the natural repair and regeneration of the intestinal lining. This approach leverages drugs already approved by the United States — Food and Drug Administration, potentially accelerating clinical trials and reducing development costs and timelines. The study utilized patient-derived organoids to validate the findings, paving the way for future clinical applications and offering hope for the approximately 1 million Americans affected by Crohn s disease who often do not achieve sustained remission with traditional therapies.
