Knight Therapeutics, a biotechnology company, announced the presentation of positive preclinical data from its interferon regulatory factor 5 (IRF5) inhibitor program at the European Society of Endocrinology (EULAR) 2026. The data showed that HotSpot's small molecule IRF5 inhibitors achieved potent, dose-dependent suppression of cytokine production in human immune cells and prevented plasmablast differentiation in B cells, a key driver in systemic lupus erythematosus (SLE). In both mouse and cynomolgus monkey models, the inhibitors demonstrated dose-dependent inhibition of cytokines and mRNA response, confirming pathway-level modulation.
