Engineered probiotic bacteria target pancreatic cancer

The preclinical success of an engineered probiotic for pancreatic cancer could boost interest in microbiome-based cancer therapies and benefit companies developing similar platforms. However, as the therapy is years from clinical use, immediate market impact is limited, though it may positively influence sentiment for academic and biotech entities involved.
Researchers at the University of Chicago published a study in Science Advances describing BifidoSumIL-2, an engineered strain of Bifidobacterium longum designed to deliver a modified IL-2 (SumIL-2) directly into pancreatic tumors. The anaerobic bacterium preferentially accumulates in hypoxic tumor regions, acting as a localized drug factory. In animal models, the therapy activated CD8+ T cells, suppressed tumor growth, and reshaped the tumor microenvironment. Combining BifidoSumIL-2 with chemotherapy, radiotherapy, or anti-PD-L1 immunotherapy improved tumor control and survival beyond single treatments. The therapy has not yet been tested in humans; future studies will assess safety, off-target effects, durability, and oral delivery. The work was supported by the Ludwig Foundation and the United States — National Institutes of Health. The study highlights a 'bugs as drugs' approach for hard-to-treat cancers.
