A large observational study by UCLA Health, published in JAMA Network Open, found that GLP-1 receptor agonists are associated with a 21% lower risk of fragility fractures in adults with type 2 diabetes compared to DPP-4 inhibitors. The study analyzed electronic health records of nearly 134,000 U.S. adults aged 50-90 over three years, with the strongest reductions in hip, femur, and spine fractures. The protective effect was observed only in diabetic participants, not in a matched non-diabetic cohort. The findings address concerns that rapid weight loss from GLP-1s might weaken bones. However, the study is retrospective and observational, so it does not prove causation; prospective trials are needed. The research was led by Dr. Christopher Hamad and involved collaborators. The study adds to evidence that GLP-1 drugs may have bone-protective benefits beyond glucose control and weight loss.
